Paratope mapping

In Paratope mapping, it is the antibody sequence that is subjected to all single mutations. All the variants are expressed on the surface of cells and the affinity of each of them for one or several target antigens (human, murine, simian, etc.) can be evaluated.

This approach allows to :

  • To establish precisely the paratope mapping of the antibody, to the nearest amino acid.
  • To know all the mutations that are permitted and that will allow in a second time to proceed in conscience to a Lead optimization, a simultaneous optimization of different parameters of the antibody (affinity, selectivity, cross-reactivity between species, humanization, de-immunization, specificity and stability).

The time required for the Paratope mapping is in the order of 4 to 6 weeks.
